News on Cialis StudiesAugust 24, 2007 - UroToday.com – Recently conducted studies provided a new portion of evidences as to the safety and efficacy of tadalafil use in men of all ages, who suffer from erectile dysfunction. Tadalafil is the main working ingredient of Cialis, one of the first-line treatment options for people with ED. Tadalafil belongs to the class of medications, known as PDE-5 inhibitors. UroToday.com, an online database of qualified and professional information concerning men’s health, has recently published several publications with the results of the clinical studies on tadalafil, conducted in Spain, UK, Germany, and the USA. The study, conduced in the University of Navarra, Spain, was aimed at characterizing the tadalafil pharmacokinetics and finding out the potential factors, which may influence it. In simple words, drug pharmacokinetics shows what happens to a particular medication once it is delivered to a human body. For example, pharmacokinetics shows how fast the medication is absorbed into the bloodstream, and how much time it takes for a human body to eliminate the medication out of the system. Different population models were designed by scientists to reflect average patient profile. In total 227 persons with mild to severe erectile dysfunction participated in the study. All the patients were divided into groups and were treated with either 2-, 5-, 10-mg dose of tadalafil or placebo (sugar pill). The study showed that tadalafil pharmacokinetics was in linear dependence with the dose and duration of treatment. The scientists proved the fact that tadalafil may start working very quickly after oral administration, because of its very rapid absorption rate. Furthermore, the efficacy of this medication was not influenced by such factors as age, weight, smoking status, alcohol consumption, liver enzyme status, ED severity, cardiovascular condition, or diabetes mellitus. Another study, which was also conducted in Spain, evaluated weather the age of a person, treated with tadalafil, could affect the patterns of his sexual activity. All in all 4262 men with ED participated in the study. One group of them took tadalafil on-demand (before the planned sexual activity), and another group received tadalafil three times per week irrespective to sexual activity. The average number of sexual attempts per week decreased from 2.6-2.8 at age < or = 40 years to 1.9 at age >70 years. However, age did not significantly influence the time of sexual activity after dosing. Most people started their sexual activity late in the evening in approximately 4 hours after taking the dose of tadalafil. Interesting to note that tadalafil remained affective for up to 36 h after dosing in all age groups. However, the rate of successful attempts decreased with age (75% at age < 60 years, 68% at age 60-70 years, and 60% at age > 70 years). The study also showed that tadalafil was well tolerated in patients of all ages. The observational cohort study, conducted in the UK, showed that the use of tadalafil in order to improve erectile performance did not affect the occurrence of cardiovascular events, such as heart attack, chest pain or myocardial infarction. Analyzing data from several studies, the scientists of the Drug Safety Research Unit, UK, noted that the risks to develop heart disorder in people receiving tadalafil (Cialis) were similar to the risks of general male population. Speaking about the influence of PDE-5 inhibitors, such as tadalafil (Cialis), vardenafil (Levitra), and sildenafil (Viagra), on the human cardiovascular system, the scientists from the Virginial Commonwealth University note that these medications may even have protective effect on the heart. Based on the modern researches and clinical studies of PDE-5 inhibitors, it is very likely that soon that class of medications will be prescribed not only for treating erectile dysfunction, but also for preventing hypertension, endothelial dysfunction, ischemia/reperfusion injury, myocardial infarction, and heart failure.
